Effect of Differentiation Inducers on the Production of NO in the Transformed Macrophages

Abstract

The effect of treatment with differentiation inducers such as retinoic acid, 1, 25-dihydroxyvitamin D3 (DHVD3) or 12-O-tetradecanoyl phorbol-13-acetate (TPA) on the production of NO was analysed the transformed macrophages activated by combined
interferon-¥ã(¥ãIFN) and lipopolysaccharide (LPS).
The production of NO was increased in the transformed cells by treating with combined ¥ã IFN and LPS compared with each agent-treated cells. The production of NO in the activated transformed cells was suppressed by the addition of
NG-monomthyl-L-arginine (NMLA) or dexamethasone.
The addition of retinoic acid, DHVD3 or TPA didn't affect the production of NO in the normal macrophages treated with comined ¥ãIFN and LPS. The production of NO in the activated transformed cells was decreased by adding retinoic acid. That
wasn't
changed by adding DHVD3, while being increased by adding TPA.
These results suggest that the production of NO was decreased by the additon of retinoic acid to the transformed macrophages, while being increased by the additon of TPA.
